OVERVIEW

A gender reveal blood test is performed during pregnancy, from as early as 7 weeks, and works by analysing tiny fragments of the baby's DNA present in the pregnant person's bloodstream. It's specifically designed to predict whether the fetus is likely male or female.

Although the test can technically be done from 7 weeks, we recommend waiting until at least 8 weeks. This is because factors beyond gestational age can influence how much baby DNA is present in the blood. If the amount of baby DNA is too low, the test may fail to detect the Y-chromosome, which could lead to an inaccurate result.

  • If Y-chromosome DNA is in your blood, the baby is a boy.
  • If there is no male DNA, the baby is a girl.

GENDER REVEAL BLOOD TEST

A gender reveal blood test is a blood test performed during pregnancy from 7 weeks that analyses small fragments of the baby's DNA circulating in the pregnant person's bloodstream. The test is designed specifically to predict whether the fetus is likely to be male or female.

This is a highly sensitive DNA test – 99% Accurate.

While this test can be done from 7 weeks pregnant, we do recommend doing the test from 8 weeks as there are other factors, other than how many weeks pregnant which can affect the amount of baby DNA in the blood. If there is insufficient baby DNA in the blood there is a chance the test will not detect the Y-chromosome and the result could be wrong.

Here are some of the most frequently asked questions about the Gender Reveal Blood Test.

How does the test work?

In pregnancy, the placenta releases tiny pieces of baby DNA (circulating cell-free foetal DNA) into your bloodstream. Our lab checks your blood sample for DNA from the Y-chromosome, found only in boys. If found, you’re having a boy. If it’s not found, you’re having a girl.

Unlike most of our DNA, found inside a cell’s nucleus, when a woman is pregnant tiny pieces of the DNA from her developing baby are present in her bloodstream. These fragments are free in the circulation and called cell-free DNA (cfDNA). Our sex chromosomes determine whether we are male or female. Females have 2 X-chromosomes (XX) and males have an X-chromosome and a Y-chromosome (XY). As a woman has only female sex chromosomes in her blood, if we analyse the blood of a pregnant woman and find a Y-chromosome then we can assume the baby she is carrying is a male. 

Does the test detect any abnormalities?

The test cannot identify a possible sex chromosome abnormality. Sex chromosome abnormalities are very uncommon but could affect the result. 

We offer two tests that do screen for chromosomal conditions - NIPT3 and NIPT5

PrenatalSafe NIPT 5 and NIPT3 are  safe, accurate and non-invasive prenatal screening tests, available privately in the UK for women who are at least 10 weeks pregnant.

This advanced blood test screens for three (NIPT3) or five (NIPT5) key chromosomal conditions in the baby without posing any risk to the pregnancy. 

How accurate is this test?

99%. Baby DNA in mum’s blood comes from the placenta (the organ that connects the baby to the mum). 99% of the time this is identical to the baby’s sex. 

Unlike other early baby gender tests, this test looks at more than one area of DNA on the Y-chromosome. Using TWO Y-chromosome DNA markers reduces the effect that any variation in one of these Y-chromosome markers could have on the accuracy of the test and increases sensitivity for male DNA. 

Please note that research suggests mum’s weight can affect how much foetal DNA is in her blood, due to the increased blood volume. Mum’s with a BMI over 30 should only do the test after 8-10 weeks pregnant, as there is a slightly higher chance that there will not be enough male DNA in her blood to be detected between 7 – 8 weeks.

The test cannot protect against Vanishing Twin Syndrome (VTS). VTS is a miscarriage that occurs in a multiple pregnancy (such as twins or triplets), where one embryo stops developing and is reabsorbed. It is most common in the first trimester, often leaving a single, healthy surviving baby to develop to full term. Because the vanished twin's tissue and gestational sac break down and are absorbed by the mother, the surviving fetus or the placenta, the pregnancy continues for the surviving baby. VTS happens in the first trimester. It is difficult to know how often it occurs as many are never seen on scan. It is unknown how long the placenta will release that baby’s DNA and therefore in a very small number of cases a false male result could occur. By the time a 12 week scan is conducted there is no evidence of the other twin at all. 

What stage of my pregnancy can I take the test?

You must be at least 6 weeks pregnant. Testing earlier means the foetal DNA in your blood will be too low for a result. If your cycles are irregular, take an early scan to confirm your dates so you test at the right time.

We do, however, recommend doing the test from 8 weeks as there are other factors, other than how many weeks pregnant which can affect the amount of baby DNA in the blood. If there is insufficient baby DNA in the blood there is a chance the test will not detect the Y-chromosome and the result could be wrong. 

Does my high BMI impact the test?

A BMI 30+ is linked to low levels of foetal DNA in your blood. We recommend an early scan confirming you’re at least 7 weeks pregnant (ideally 8) with a visible baby heartbeat before you test for accurate results.

Does my past pregnancy / miscarriage impact results?

No. Foetal DNA clears from your bloodstream within 48 hours after birth or a miscarriage. Test not impacted.

I'm pregnant with non-identical twins, can you confirm both genders?

No. Your babies might be different sexes. A positive Y-Chromosome means at least 1 baby will be a boy.

I'm pregnant with identical twins, can you confirm both genders?

Yes. Your baby's DNA is nearly identical. A positive Y-Chromosome = 2 baby boys. Negative = 2 baby girls.

My result says 'no Y-Chromosome', what does it mean?

If no Y-Chromosome is present in your blood that means no male babies are present, you are having a female baby.

My result is inconclusive, what does it mean?

That your sample didn’t contain enough blood and placental DNA to give a result. Or if you tested too early, have high lipid (fat) levels in your blood, or are an older mum, or in IVF pregnancies where the placenta develops later. Simply, repeat the test 2 weeks later to allow time for DNA levels to rise for a clear result.

What if the ultrasound shows something different?

In this situation the only way to really know the baby’s gender is waiting until birth. There are two reasons why they may differ;

  1. The ultrasound relies on looking for a penis or a vulva, alongside other subtle changes. These can be highly dependent on the position of the baby at the time of the scan. Whilst scans have improved over the years they are still much more subjective than DNA.
  2. The DNA test result is incorrect as not enough DNA of the baby was present in mum’s blood at the time the blood was taken. See “how accurate is the test” for more information.

Can I do the test if I am still breastfeeding?

Yes. Breastfeeding does not affect the test results. The Gender Reveal test looks for male chromosomes in mum’s blood. Breastfeeding is a hormonal change, which has no impact on genetics, and therefore it has no impact on the Gender Reveal test accuracy or results.

Can the Gender Reveal test be used as a pregnancy test?

Gender Reveal cannot confirm a pregnancy. The test is only looking for male DNA in the blood. You must confirm your pregnancy with a doctor before doing the Gender Reveal test. It is essential to know dates of mum’s last period before doing the test.

Does Gender Reveal Test replace an ultrasound?

No, ultrasounds are essential to monitor a baby's development during the pregnancy.
